چکیده

A thermophilic fungal strain producing both pectinase and polygalacturonase was isolated after primary screening of 120 different isolates. The fungus was identified as Aspergillus fumigatus Fres. MTCC 4163. Using solid-state cultivation, the optimum levels of variables for pectinase and polygalacturonase (PG) production were determined. Maximal levels of enzyme activities were achieved upon growing the culture in a medium containing wheat bran, sucrose, yeast extract and (NH4)2SO4 after 2-3 days of incubation at a temperature of 50oC. Highest enzyme activities of 1116 Ug for pectinase and 1270 Ug for polygalacturonase were obtained at pH 4.0 and 5.0, respectively.

Production, Purification, and Characterization of Polygalacturonase from Rhizomucor pusillus Isolated from Decomposting Orange Peels

A thermophilic fungal strain producing polygalacturonase was isolated after primary screening of 40 different isolates. The fungus was identified as Rhizomucor pusilis by Microbial Type Culture Collection (MTCC), Chandigarh, India. An extracellular polygalacturonase (PGase) from R. pusilis was purified to homogeneity by two chromatographic steps using Sephadex G-200 and Sephacryl S-100. The pur...

Biosynthesis of Industrially Relevant Extracellular Pectinase from Aspergillus terreus NCFT 4269.10 Using Orange Peels as Substrate

In this study, Aspergillus terreus NCFT4269.10 was selected for the biosynthesis of pectinase after screening from fifteen native fungal strains as the performance of A. terreus was best in pectin hydrolyzing activity (2.75 cm). Aiming at the cost effective fermentation, several agro-industrial residues such as, Pearl Millet (PM), Finger Millet (FM), Orange Peels (OP), Mustard oil Cake (MoC) an...

Pectinase Production by Penicillium Viridicatum Rfc3 by Solid State Fermentation Using Agricultural Wastes and Agro-industrial By-products

Pectin lyase and polygalacturonase production by newly isolated Penicillium viridicatum strain Rfc3 was carried out by means of solid state fermentation using orange bagasse, corn tegument, wheat bran and mango and banana peels as carbon sources. The maximal activity value of polygalacturonase (Pg) (30U.g) was obtained using wheat bran as carbon source while maximal pectin lyase (Pl) (2000 U.g)...

Biochemical Characterization, Thermal Stability, and Partial Sequence of a Novel Exo-Polygalacturonase from the Thermophilic Fungus Rhizomucor pusillus A13.36 Obtained by Submerged Cultivation

This work reports the production of an exo-polygalacturonase (exo-PG) by Rhizomucor pusillus A13.36 in submerged cultivation (SmC) in a shaker at 45°C for 96 h. A single pectinase was found and purified in order to analyze its thermal stability, by salt precipitation and hydrophobic interaction chromatography. The pectinase has an estimated Mw of approximately 43.5-47 kDa and optimum pH of 4.0 ...
